Orodispersible tablets in modern pharmaceutics: Formulation, evaluation, and clinical relevance

Abstract: Tablets are one of the oldest and most commonly employed dosage forms in the pharmaceutical industry and due to high patient compliance, the oral route of administration is the most preferred route of administration. Although conventional solid dosage forms are highly efficacious and offer great patient compliance but they lack majorly due to high first pass metabolism and thus they are unable to provide immediate action. Also, some categories of patients such as pediatric, geriatric and dysphagic patients find it difficult to administer conventional tablets and thus orodipsersible tablets can offer great advantages such as no need of water, rapid action due to fast dissolution in mouth. Orodispersible tablets (ODTs) are the new generation solid dosage forms or one advancement in tablets, which allows tablets to dissolve immediately upon administration. This is mainly due to the presence of superdisintegrants which allows rapid dissolution of tablets in the mouth. Now-a-days ODTs are widely used as they improve bioavailability, increase patient compliance and caters need of targeted patients. In this review article, comprehensive information on orodispersible tablets including its advantages over conventional dosage forms, mechanism of rapid action, formulation, manufacturing methods, patented technologies and patents granted, challenges, future prospects, ongoing and completed clinical trials along with marketed formulations is given.
